SARS-CoV-2 emerged through natural zoonotic spillover, most likely at the Huanan Seafood Wholesale Market in Wuhan in late 2019. The WHO-China Joint Study (March 2021) ranked a laboratory leak as 'extremely unlikely.'

The DEFUSE proposal — submitted to DARPA's PREEMPT program in March 2018 by EcoHealth Alliance (Peter Daszak) with the Wuhan Institute of Virology (Shi Zhengli) and the University of North Carolina (Ralph Baric) — proposed inserting human-specific furin cleavage sites into spike proteins of SARS-related bat coronaviruses for vaccine-development testing. DARPA rejected the proposal on biosafety grounds; the full document was released by DRASTIC researchers via FOIA in September 2021.
The Wuhan Institute of Virology's viral-sequence database — containing approximately 22,000 entries on bat coronaviruses including unpublished sequences — was taken offline on 12 September 2019, several weeks before the earliest documented Wuhan cases. WIV director Shi Zhengli has stated the database was removed due to 'hacking attempts'; the database has never been restored.
The Bruttel, Washburne, and VanDongen preprint 'Endonuclease fingerprint indicates a synthetic origin of SARS-CoV-2' (October 2022, BioRxiv) identified a statistically unusual pattern of restriction enzyme recognition sites (BsaI, BsmBI) in the SARS-CoV-2 genome consistent with a no-see'm cloning strategy used in Ralph Baric's published work; the paper was heavily contested but not retracted.
Three Wuhan Institute of Virology researchers — reported by U.S. intelligence and first published by the Wall Street Journal on 23 May 2021 — sought hospital treatment for COVID-like respiratory illness in November 2019, before the earliest officially acknowledged Wuhan cases. The WSJ later (June 2023) named the three: Ben Hu, Yu Ping, and Yan Zhu.
On 26 February 2023 the Wall Street Journal reported that the U.S. Department of Energy — which oversees the U.S. national-laboratory complex including biosafety research — had updated its formal assessment to conclude that a lab leak was the 'most likely' origin of SARS-CoV-2, joining the FBI (which had reached the same 'moderate confidence' conclusion in 2021). The DOE's underlying classified analysis has not been publicly released.

Worobey, Pekar, et al. ('The Huanan Seafood Wholesale Market in Wuhan was the early epicenter of the COVID-19 pandemic,' Science, July 2022) used spatial epidemiology of the earliest 174 confirmed December 2019 cases to show the cases cluster within meters of the wildlife-section stalls at the Huanan market, not near the WIV (which is approximately 12 km away).
Pekar, Magee, et al. ('The molecular epidemiology of multiple zoonotic origins of SARS-CoV-2,' Science, July 2022) used phylogenetic reconstruction to show that both major early SARS-CoV-2 lineages (A and B) circulated at the Huanan market in December 2019, consistent with two separate spillover events rather than a single lab-released strain.
Multiple intermediate-host coronaviruses (raccoon dogs, civets, masked palm civets) were documented to be sold live at the Huanan market in late 2019 by Xiao et al. (Scientific Reports, June 2021); environmental swabs from the wildlife stalls subsequently tested positive for SARS-CoV-2 RNA.
No published WIV viral sequence (including RaTG13, sampled 2013 from Mojiang miners' cave) is closer than ~96% to SARS-CoV-2 — too distant in evolutionary terms to be a direct engineered ancestor; the gap implies an unknown intermediate animal host that would be required regardless of origin theory.
The furin cleavage site itself (PRRA insertion) has natural analogs in other coronaviruses (MERS-CoV, HKU1, IBV) and is not by itself an unambiguous fingerprint of laboratory engineering; the Andersen et al. proximal-origin paper (Nature Medicine, March 2020) made this argument — though Andersen's contemporaneous private Slack messages (released by House Oversight, July 2023) showed substantially more openness to a lab origin than the paper acknowledged.
